I followed iFixit instruction up to step 21st (link: https://www.ifixit.com/Guide/iBook+G4+12-Inch+1.33+GHz+Hard+Drive+Replacement/703) I have concern that remove those adhesive cable. Is it really necessary for swapping hard drive?

The guides are usually a bit more intense than they need to be as they are usually taken from a full tear down guide. I don't see any impediment as to why that DC Jack connector has to be remove as far as replacing the Hard Drive is concerned.
